    Chromatin dynamics during DSB repair

    We show that double strand breaks (DSBs) induced in chromatin of low as well as high density by exposure of human cells to ?-rays are repaired in low-density chromatin. Only slight movement of sporadic DSB loci for short distances was noticed in living cells associated with chromatin decondensation around DSBs. This frequently resulted in their protrusion into the low-density chromatin domains. In these regions, occasional clustering of DSB foci was seen that resulted in lesions repaired only with difficulty.

    Dvouřetězcové zlomy DNA (DSB) vyvolané v řídkém i hustém chromatinu po expozici lidských buněk gama záření jsou reparovány v řídkém, dekondenzovaném chromatinu. Dekondenzace chromatinu v okolí DSB byla doprovázena pouze méně výrazným pohybem sporadickýchDSB. Tento pohyb často vedl k jejich protrusi do jaderných domén s nízkou hustotou chromatinu. V těchto doménách pak docházelo k občasnému klastrování DSB ohnisek, což vedlo ke vzniku jen těžce opravitelných lézí DNA.
