    Inhibitors of NADPH oxidase decrease endotoxin mediated induction of inducible nitric oxide expression in mouse macrophages

    Intracellular reactive oxygen and nitrogen species (RONS) produced by NADPH oxidases and NOS are suggested to be important mediators augmenting the response of cells to endotoxin. Through this mechanisms NO could positively promote NOS expression and further increase of NO production. The aim of this study was to characterize modulation of endotoxin activated macrophage NO production by the inhibition of RONS producing NADPH oxidases and NOS.

    Inhibitory NADPH oxidázy snižují endotoxinem zprostředkovanou indukci exprese inducibilní syntázy oxidu dusnatého u myších makrofágů

    Stimulace produkce NO makrofágy aktivovanými endotoxinem vyžaduje tvorbu RKDM závislou na NADPH oxidázách a iNOS. Inhibice produkce NO makrofágy pomocí DPI není způsobeno pouze inhibicí aktivity iNOS, ale snížením exprese tohoto enzymu, což potvrzuje roli RKDM v aktivaci makrofágů endotoxinem. Lze tedy konstatovat, že modulace intracelulární tvorby RKDM může mít farmakologické uplatnění při léčbě plicního zánětu spojeného s expozicí endotoxinem.
