The aryl hydrocarbon receptor (AhR) is a transcription factor belonging to the basic helix-loop-helix Per-ARNT-Sim family of transcriptional regulators and a major regulator of xenobiotic metabolism. AhR and its target genes have been found to be deregulated in a number of cancer types, which has been linked with cancer development and progression. Nevertheless, upregulation of several xenobiotic-metabolizing enzymes and transporters via AhR signaling, which is observed in some cancer types, also has the potential to alter the metabolism and efflux of anticancer drugs. Herein, we provide a short overview of the current understanding of the potential impact of deregulated AhR expression and activity on drug metabolism in cancer cells. We focus primarily on established AhR gene targets, including phase I enzymes such as cytochrome P450 family 1 enzymes. Furthermore, this chapter is intended to provide a broader overview of the impact of AhR signaling alterations that have been uncovered in tumor cells.
