Animals lacking link protein have attenuated perineuronal nets and persistent plasticity
Original language description
Chondroitin sulphate proteoglycans in the extracellular matrix restrict plasticity in the adult central nervous system and their digestion with chondroitinase reactivates plasticity. In this article has been measured the changes in the CSPGs that occur during the critical period in rats and mice. The molecular event that triggers PNN formation has been identified, showing that the PNN component cartilage link protein Crtl1 (Hapln1) is up-regulated as the structures form. In keeping with the hypothesis that PNN maturation down-regulates plasticity, we found that adult plasticity in the visual and somatosensory systems of mice lacking Crtl1 was strongly enhanced. Overall, our findings demonstrate that the formation of PNNs triggered by neuronal Crtl1 synthesis is a key event in the diminuation of plasticity in the adult CNS and suggest that modification of PNNs is how CSPGs reactivate plasticity.
