Original language description
Controlled production of granulocytes in health and disease is a criticalnprocess in hematopoiesis. Despite extensive investigation of the cellularnmachinery that controls blood cell production, the mechanisms regulatingngranulopoiesis are still unclear. It has been shown that thenCCAAT/enhancer-binding protein (C/EBP) family of transcription factorsnplays a critical role in this process, however the function of C/EBPg innnormal and emergency granulopoiesis remains still elusive. Here, wengenerated a C/EBPg conditional knockout (KO) mouse model where Cebpg isndepleted specifically in the hematopoietic system. We observed that CebpgnKO mice are viable, and present no signs of disease. Consistently, wenshowed that production of mature blood cells as well as function ofnhematopoietic stem and progenitor cells remains unaffected upon Cebpgnablation. Next, we investigated the role of this transcription factornduring stress granulopoiesis, induced by administration of LPS, G-CSF,nCandida albicans, or cyclophosphamide. Surprisingly, challenging Cebpg KOnmice with these agents resulted in an emergency granulopoiesis responsensimilar to the one exhibited by WT control mice. Altogether, our resultsndemonstrate that the transcription factor C/EBPg, although being expressednat high levels in all hematopoietic cells, is dispensable for steady-statenand emergency granulopoiesis.n
