    Whole-cell biosensor- Influence of optical fiber element shape on detection of low-level bioluminescence

    The whole-cell biosensor which used a special optical fiber element (OFE) from pure silica with an active layer contents bioluminescent bioreporters was developed as a real in-situ detector. The sensitivity depends on a value of detected bioluminescence(BL), which can be very low. In this study, we explore the influence of OFE shape on BL transition as well as number of cells immobilized into the active layer on detecting low-level bioluminescent signals. The bioluminescence signal was produced by thegenetically engineered bacterial bioreporter (light source). Mathematical model of OFE shape was developed based on geometric optics. Four different shapes of OFE were studied experimentally. For development of mathematical model of the optimal OFE shapefrom point of view maximum detected intensity of BLwas applied parameters of the best OFE from experimental measurements.
