Enzymatic glycosylation of phenolic and polyphenolic substances.
Project goals
The diglycosidase a-rhamnosyl-b-glucosidase from Acremonium and/or from other fungal sources will be purified and characterized from the proteomic point of view. a-Rhamnosidase from fungal producers will be cloned to be used as alternative to the diglycosidases for the synthetic reactions. These enzymes will be sequenced and cloned and expression in heterologous producer will be attempted. a-Rhamnosyl-b-glucosidase and a-rhamnosidase will be used for transglycosylation of various phenolic acceptors fromgroup of flavonoids (silybin), polyphenols (e.g., resveratrol) and some other phenolic and aromatic precursors. The biological activity of the compounds carrying 1 or 2 glycosidic residues will be tested. We will test mainly antioxidant activities within vitro tests (DPPH) and ex-vivo tests (microsomal oxidation).

Diglykosidasa a-rhamnosyl-b-glukosidasa z kmenů hub Acremonia a z dalších fungálních zdrojů bude purifikována a charakterizována z proteomického hlediska. a-Rhamnosidasa z fungálních producentů bude klonována a dále využita jako alternativa k výše uvedeným diglykosidasám pro synthetické reakce. Tyto enzymy budou sekvenovány a testována exprese v heterologních producentech. a-Rhamnosyl-b-glucosidasa a a-L-rhamnosidasa bude použita pro transglycosylace různých fenolických akceptorů ze skupiny flavonoidů (silybin), polyfenolů (např. resveratrol) a dalších fenolických a aromatických substrátů. Biologická aktivita těchto látek nesoucích 1 - 2 nové glykosidické zbytky bude testována především jako antioxidační aktivita v in vitro testech (DPPH) a ex-vivo testy (mikrosomální oxidace).
