    The major objective of this project is to decipher the structural basis of the interaction between the calmodulin and the N-terminal actin binding domain of plectin isoform 1a and to provide the model with a functional-structural basis. Following goals will be pursued: i) To investigate interaction of calmodulin with the N-terminal domain of plectin isoform 1a (1aABD) - structural analysis of the interaction between calmodulin and N-terminal domain of plectin isoform 1a ii) To study the mechanism of calmodulin-dependent disruption of the interaction between plectin and integrin - analysis of the disassembly mechanism of intergrin-plectin complex after binding of calmodulin iii) To address the role of calmodulin in modulation of the function of actin binding domains - analysis and comparison of ABDs from a selected number of actin binding proteins, which calmodulin and CaM-like domain bind. iv) To test and validate proposed model of calmodulin-plectin 1a interaction ex vivo - structural analysis of theinteraction between calmodulin and N-terminal domain of plectin isoform 1a and analysis of the disassembly mechanism of intergrin-plectin complex after binding of calmodulin ex vivo.
