Modifikace proteinů reaktivními aldehydy odvozenými z metabolismu polyaminů

  • Annotation in Czech

The aim of this study is to evaluate the impact of polyamine-derived aminoaldehydes on living cells, focusing on the induced protein modifications, their characteristics, and extent. We plan to treat model and real protein samples with the aminoaldehydes APAL, AcAPAL, and BOPBD. These compounds can be obtained commercially (APAL) or synthesized in the laboratory: AcAPAL via acetylation of APAL, and BOPBD either through enzymatic spermine oxidation using purified pea seedling diamine oxidase or by chemical oxidation of spermidine. These compounds will serve as reagents for experimental treatments. The model proteins will include serum albumin, hemoglobin, and other selected proteins including enzymes. Real protein samples will be derived from rat brain lysates and protein extracts from microorganisms, including bacteria (e.g., Escherichia coli), yeast (Saccharomyces cerevisiae), and cyanobacteria (Synechococcus sp.). Additionally, we will use hepatocytes and rat brain cells as protein sources. Microbial, hepatic, or rat brain cells will also be directly treated with the reactive compounds at different concentrations, and the resulting protein extracts or tissue samples will be analyzed.
