Generation of temperature sensitive cell division cycle mutants in green alga Chlamydomonas reinhardtii

Project goals

Cell cycle study has been one of the fastest evolving areas in today's biology. Most of what we know about the cell cycle regulation is thanks to the production of conditional cell division cycle mutants generated in 1970s by Leland Hartwell in Saccharomyces cerevisiae and Paul Nurse in Schizosaccharomyces pombe. Here, we propose to generate a temperature sensitive cell division cycle mutants in the green alga Chlamydomonas reinhardtii, a model plant single celled organism. The aims of the proposal are:1) produce temperature sensitive cell division cycle mutants of Chlamydomonas reinhardtii, 2) identify cell cycle phenotypes and execution points for the mutants, 3) form complementation groups, 4) characterize in more detail one to two of the most interesting mutants.
