    Targeting NOX4 in Multiple Organs: A Strategy to Combat Type 2 Diabetes Development

    Type 2 diabetes (T2DM) is a systemic metabolic disease characterized by chronic hyperglycemia and oxidative stress that drives progressive multi-organ dysfunction. This project targets NOX4, a constitutively active NADPH oxidase isoform, as a key regulator of redox imbalance in obesity-driven T2DM. We aim to elucidate the impact of NOX4-ROS signaling on the function of pancreas, adipose tissue, heart, and bone during obesity-induced T2DM development using pharmacological approaches and knockout models with respect to sex differences. In parallel, we will investigate how NOX4 signaling interacts with RNA methylation (m6A/m6Am) – an emerging epitranscriptomic mechanism implicated in metabolic stress responses. By integrating redox biology with RNA epigenetics, this project will reveal tissue- and sex-specific pathways contributing to T2DM onset and complications. The findings will identify critical windows for NOX4-targeted therapy and provide mechanistic insight into transcriptomic plasticity as a driver of disease progression.
