Uncovering electrophysiological remodeling in adult congenital heart diseases using a rat model of neonatal pressure overload
Annotation in Czech
With the remarkable success in surgical repair, patients with congenital heart disease (CHD) more frequently reach adulthood. However, our knowledge about long-term consequences of the altered hemodynamic conditions observed in both unrepaired and repaired CHD is limited. The aim of this project is to comprehensively characterize the phenotype progression of CHD with elevated left ventricular workload as one of the most frequent CHD. Using our model of rat abdominal aortic constriction from the neonatal period (neo-AAC), we will focus primarily on electrophysiological remodeling with proarythmogenic substrate formation since this is known adverse consequence of the increased workload in adults. We will characterize the molecular level, tissue characteristics, and heart metabolism that underlie the neo-AAC electrophysiological outcomes in adult rats. Further, we will address the role of sex-specific characteristics and angiogenesis in these animals. The results will enhance our understandying of electrical remodeling resulting from long-tem increased workload
