    Architecture and evolution of protein synthesis apparatus in plastids

    A plastid is an endosymbiotic organelle that has retained (reduced) genome and the machinery for its expression, including ribosomes (plastoribosomes). The structure of the plastoribosome has only been studied in a land plant (spinach). Here, we integrate various experimental and bioinformatic approaches to study the diversity of plastoribosomes across eukaryotes. Using cryo-EM, we will determine plastoribosome structures from three distantly related species, chosen for their evolutionary and socioeconomic significance. In two other species, we will identify plastoribosomal proteins through tagging and IP-MS or by spatial proteomics. Bioinformatic analyses will map plastoribosomal protein distributions across plastid-bearing groups, revealing evolutionary history, inter-ribosomal protein transfer, and past endosymbiotic events. Our findings will shed light on the evolution of the plastoribosome and complex plastids, enable comparisons with mitoribosomes, and may provide a basis for optimizing biotechnological production.
