    Impact of mTOR signaling modulation in NG2-expressing cells on the neuroregeneration after ischemic brain injury

    The present project aims to evaluate the impact of mTOR signaling modulation on the proliferation and differentiation of NG2 glia, oligopotent cells involved in brain repair after ischemic stroke, and on their contribution to post-ischemic tissue regeneration. Our goal is to investigate the cellular and molecular mechanisms that may counteract damage induced by focal cerebral ischemia (FCI) and support neuroregeneration. Our preliminary data indicate that mTOR signaling is upregulated in NG2 glia in the ischemic mouse brain. Therefore, we will use transgenic animals to inhibit or activate mTOR signaling specifically in NG2 glia, in an animal model of FCI induced by permanent occlusion of the middle cerebral artery. The effect of mTOR signaling modulation on the properties of NG2 glia at the cellular level and in vivo will be evaluated by gene expression profiling, proteomics, functional analyses, and behavioral testing. Since no broadly applicable treatment for stroke exists, we believe that our findings may help identify new directions for glia-oriented therapeutic strategies.
