    Using Reverse Genetics in Amphioxus to Inform the Development and Evolution of Chordates

    As a representative of a basal lineage within the chordate phylum, amphioxus retains many characteristics thought to have been present in the common chordate ancestor. By studying amphioxus, we can thus infer how key vertebrate traits evolved over time. Most of the current insight into vertebrate evolution using amphioxus as a model is based on descriptive gene expression studies. Here, we plan to leverage the power of CRISPR/Cas9-based reverse genetics combined with phenotypic and molecular characterization of mutants. By inactivating evolutionarily conserved genes implicated in the development of the visual system and placodes we aim to provide fundamentally novel insight into the ancestral chordate state and vertebrate-specific innovations.
