    Heart disease is the leading cause of death worldwide, with significant differences in symptoms and outcomes between males and females. Emerging evidence shows that sex chromosomes play a key role in these differences, yet the molecular mechanisms are not known. My research will bridge this knowledge gap by integrating transcriptomic, proteomic, and metabolomic approaches in genetically engineered mouse models. Using mice with genetically tagged mitochondria, we will map the molecular sex differences in healthy heart tissues, cardiomyocytes, and mitochondria, creating a more holistic model of cardiac sex differences. Leveraging a mouse model with a variable number of sex chromosomes, we will define the role of the inactive X chromosome (Xi) in sex-differential cardiac phenotypes, including mitochondrial energy metabolism. Understanding how Xi regulates these differences - providing cardioprotection to one sex while posing a risk to the other - is essential for advancing basic and clinical research, to develop more effective treatments that equally benefit male and female patients.
