This project explores the biophysical principles governing interactions between novel hybrid lipid-polymer-peptide nanocarriers and the tear film lipid layer (TFLL), aiming to improve topical drug delivery for glaucoma. Building on our recent studies of nanoscale tear film structure and dynamics, we seek to understand how nanocarrier architecture affects residence time and delivery efficiency - major limitations of current therapies. Using a combination of in vitro, ex vivo, and in silico models, we will define structure–function relationships that control carrier-TFLL compatibility. The insights gained will guide the rational design of advanced nanotherapeutics with enhanced retention at the ocular surface and more effective drug delivery.
