Mechanism of ergosterol - plant cell interaction

Project goals

Lipid substances are responsible for many signal mechanisms in plants. They play the role of hormones (brassinosteroids) or signal molecules (ergosterol). Ergosterol, the main sterol of  the fungi cytoplasmatic membrane, is one of the lipids which activate defense reaction in plants; at standard conditions it is not present in them. It triggers defense reaction in tobacco cells even in nanomolar concentration. The defense reaction is characterized by production of active oxygen species, alkalization ofextracellular medium, production of phytoalexin capsidiol and activation of pathogen related proteins expression, on the other hand, it does not lead to cell death. Thus ergosterol is suitable for protecting plants against fungi. However, the mechanism of triggering the plant defense reaction by ergosterol is not known for the present. The project will be focused on early recognition of ergosterol by tobacco cells. Moreover, the signal pathway elicited by ergosterol has not been fully explained yet. Bynow it has been found that ergosterol triggers defense reaction via activation of A2 phospholipase and C proteinkinase. Therefore, the project will be also focused on completion of the so far unexplained components of the signal pathway elicited by ergosterol (e g. release of Ca2+ from a vacuole).
