Analysis of molecular structure of the short arm of wheat chromosome 1B (Triticum aestivum)

Project goals

Analysis of the complex genome of common wheat is hampered by its enormous size and allohexaploid nature. Its fractionation into structural and functional parts, chromosomes, may simplify its mapping and analysis. BAC (Bacterial Artificial Chromosome) DNA library specific for short arm of chromosome 1B (1BS) has been constructed. This library will be used as a source of clones for detailed analysis of molecular structure of gene-rich regions on 1BS. The main goal of the project is the construction of ahigh-density cytogenetic map for 1BS and generation of chromosome-specific markers - CSTs (Chromosome Sequence Tags). The obtained data will be used for comparative studies group 1 chromosomes in other cereals.

    Analýza molekulární struktury krátkého ramene chromozómu 1B pšenice (Triticum aestivum)

    Značná velikost genomu hexaploidní pšenice a jeho allopolyploidní povaha ztěžují jeho detailní analýzu. Rozdělení genomu na jednotlivé chromozómy a jejich klonování v knihovnách dlouhých inzertů DNA otevřelo možnost studovat definované části tohoto genomu. Knihovna DNA specifická pro krátké rameno chromozómu 1B hexaploidní pšenice (1BS), klonované ve vektoru BAC (bakteriální umělý chromozóm) bude využita pro analýzu molekulární struktury genově bohatých oblastí nacházejících se na 1BS. V průběhu řešeníbude vytvořena cytogenetická mapa o vysoké hustotě s využitím nově odvozených chromozómově specifických CST (Chromosome Sequence Tag) markerů. Získané výsledky budou využity také pro studium homologních oblastí chromozómů skupiny 1 u jiných druhů
