Plant cells are defined by their cell walls, which hold the cells together, protect them, and determine their shape. While callose, a polysaccharide related to the cell wall, is not typically recognized as a "classical" component, it certainly deserves attention. Callose is synthesized and degraded dynamically and polarly to regulate essential physiological processes such as cytokinesis, plasmodesmata gating, phloem transport, pollen development, and pathogen defence. This process is enabled by one of the biggest transmembrane proteins in plants, Callose Synthases (CalS), encoded by a family of 12 genes in Arabidopsis. Although the sites of callose deposition are known, the mechanisms regulating the spatiotemporal activity of CalS remain unclear. Our preliminary data suggest that membrane lipid nanodomains, particularly those enriched in anionic phospholipids, may serve as critical platforms for the targeted localization and activity regulation of Callose Synthases. This project aims to entangle the dynamic interplay between polar callose synthesis and lipid signalling.
