    Genetika a šlechtění průmyslových mikroorganismů

    Jedná se o přípravu nových linkosamidových antibiotik u Streptomyces lincolnensis, S. roseolus a S. caelestis pomocí mutageneze a selekce a pomocí genového inženýrství. Nová antibiotika nebo jejich deriváty budou izolovány a jejich chemická struktura bude určena pomocí nukleární magnetické rezonance a hmotové spektrometrie. Kultivační podmínky pro produkci budou optimalizovány nejprve v laboratorním měřítku a v malých fermentorech. Získané výsledku budou použity v poloprovozním měřítku. Budou izovolánataková množství látek, aby mohla být testována jejich biologická aktivita. Klinické kmeny methicilin-resistentní bakterie Staphylococcus aureus budou použity jako testovací mikroorganizmy. Cílem projektu je získat nové antibakteriální látky proti rezistetním klinickým patogenním bakteriím.

    Some genes coding biosynthesis of the important antibiotic lincomycin were identified and genes and proteins with a regulatory role in biosynthesis were tested. Production Streptomyces lincolnensis strains and new lincomycin derivatives were prepared.
