    Selektivní NMR značení jako nástroj pro charakterizaci proteinových komplexů zapojených do neurodegenerativních onemocnění

    Cílem projektu je implementace fluorovou roztokovou NMR spektroskopie na Tau a Map2c protein a její využití ve třech oblastech: 1) Charakterizace efektu fosforylace na 19F NMR signály. 2) Studiu interakčních vlastností Tau a Map2c proteinů s jejich vazebnými partnery – 14-3-3 proteinem a SH3 doménou plektinu. Hlavním předmětem studia budou vazebná místa a vazebná afinita do těchto míst. 3) Monitorování oligomerizace Tau a Map2c proteinů. Stanovení kinetiky oligomerizace a oblastí největších konformačních změn.

    Protocols for 19F labeling of different aromatic residues within Tau and its binding partner 14-3-3 protein were optimized, and the individual signals within the 19F NMR spectra were assigned. Subsequently, 19F NMR spectroscopy was successfully applied for the monitoring of oligomeric and conformational changes as well as of the interaction properties. Experimental NMR data were complemented by the computational simulations.
