Pathological expansion of microsatellites in the human genome.

Project goals

1, Identification of regions in the human genome most prone to expansion by means of bioinformatic analysis. 2, PCR of these regions to assess their propensity to expansion. 3, Biophysical analysis of the expanding fragments to identify DNA secondary structures participating in the expansion. 4, Summary of the results into rules predicting expansion propensity to any nucleotide sequence. Application of the rules to the whole human genome. Identification of the human genome regions most prone to the pathological expansions.

Podrobení těchto oblastí PCR s cílem určit jejich náchylnost k patologickému prodlužování. 3. Biofyzikální analýza prodlužujících se fragmentů s cílem určit sekundární struktury DNA, které se na prodlužování podílejí. 4. Shrnutí výsledků do podoby pravidel přiřazujících náchylnost u prodlužování libovolné nukleotidové posloupnosti. Aplikace těchto pravidel na celý lidský genom. Identifikace míst vgenomu nejnáchylnějších k patologickému prodlužování.
