Original language description
Monoclonal antibody 2E12 was prepared by immunization of mice with fresh cells of chronic myeloid leukemia cell line MOLM-7. Subset of cells of 15 tested cell lines and 10 cultured patients leukemic cells was positive with the 2E12 antibody. KG-1 and HL-60 cell lines were treated by camptothecin for 4 hours, washed and further cultivated up 48 hours. A significant increase of the positivity with 2E12 was detected after 24 and 48 hours of culture of both cell lines, which well correlated with the increase of mitochondial apoptotic antigen Apo 2.7 and with the subG1 peaks. The 2E12 positive cells were morphologically in the programmmed cell death, possibly apoptosis. It is suggested, that 2E12 antibody detects an antigen on apoptotic cells, which can bepart of the signaling process for the ingestion by phagocytes.
