    A single-nucleotide polymorphism at codon 108/158 of the catechol-O-methyltransferase (COMT) gene alters the metabolism of catecholamine neurotransmitters and contributes to the etiology of several mental disorders. We present a rapid real-time PCR method for COMT genotyping via melting point analysis with fluorescent hybridization probes. 58 of 118 investigated healthy volunteers (49%) were heterozygous MetNal, 25 subjects (21 %) were homozygous Met/Met, and 35 subjects (30%) were homozygous ValNal. The estimation ofVal-108/158 allele frequency was aDout 54%. Detection ofthe genetic polymorphism at COMT codon 108/158 by real-time PCR could be an attractive approach providing rapid and accurate results of DNA analysis.

    Jednonukleotidový polymorfismus na kodonu 108/158 genu pro katechol-O-methyl transferázu (COMT) ovlivňuje metabolismus katecholaminergních neurotransmiterů a podílí se na etiologii řady duševních poruch. Je prezentována metoda rychlé PCR v reálném čase pro genotypizaci COMT cestou melting point analýzy se značením fluorescenční hybridizací. 58 ze 118 zdravých dobrovolníků (46%) byli heterozygoty pro Met/Val, 25 osob (21%) byli homozygoti Met/Met a 35 osob (30%) byli homozygoti Val/Val. Odhad frekvence výskytu Val alely je 54%. Detekce dalších polymorfismů codonu 108/158 genu pro COMT metodou PCR v reálném čase je vhodným nástrojem poskytující rychlou a přesnou DNA analýzu.
