Molecular study on acetolactate synthase gene (ALS) of two Czech biotypes of kochia (Kochia scoparia (L.) Schrad.)

  • Original language description

Two regions of the ALS structural gene known to vary in ALS-resistant biotypes were amplified, digested with restriction endonucleases and one of them sequenced for chlorsulforon-resistant and chlorsulforon-susceptible Kochia scoparia biotypes. The Jihlava resistant kochia biotype contained a single nucleotide substitution in Domain B, predicting a tryptophan (in susceptible) to leucin (in resistant) amino acid change. No other interbiotypic nucleotide differences predicted amino acid changes in the sequenced region, suggesting that the amino acid substitution reported above is responsible for resistance to ALS-inhibiting herbicides in the biotype.
