    Quantification of Pyrenophora teres in infected barley leaves using real-time PCR

    Net blotch is a barley foliar disease caused by two forms of Pyrenophora teres: Pyrenophora teres f. teres (PTT) and Pyrenophora teres f. maculata (PTM). To monitor and quantify their occurence during the growing season, diagnostic system based on real-time PCR was developed. TaqMan MGB primers and probes were designed that showed high specificity for each of the two forms of P. teres. As a host plant internal standard, TaqMan MGB primers and probe based on RacB gene sequence were designed. The method was optimised on pure fungal DNA and on plasmid standard dilutions. Quantification was accomplished by comparing Ct values of unknown samples with those obtained from plasmids standard dilutions. The assay detects down to five gene copies per reaction. Itis able to produce reliable quantitative data over a range of six orders of magnitude. The developed assay was used to differentiate and quantify both forms of P. teres in infected barley leaves.     Síťová skvrnitost je listové onemocnění ječmene. Je způsoveno dvěma formami houby Pyrenophora teres: Pyrenophora teres f. teres (PTT) a Pyrenophora teres f. maculata (PTM). Aby bylo možno monitorovat výskyt této houby v průběhu vegetace, byla vyvinuta metoda její rychlé a spolehlivé detekce pomocí PCR v reálném čase. Byly navrženy a testovány specifické TaqMan sondy a sady primerů. Sety rozlišují obě formy a detekují je zvlášť v multiplexovém uspořádání real-time PCR. Vyvinutá metodika byla použita k detekci a stanovení P. teres ve vzorcích jak po umělé infekci, tak i pro monitoring výskytu v polních podmínkách. Byla ověřena vysoká specifita reakce a její citlivost. V polních podmínkách v oblasti Kroměříž byl takto zjištěn vyšší obsah patogena Pyrenophora teres f. teres v obou letech sledování 2003 a 2004. Vyvinutá metoda je důležitým nástrojem pro monitorování výskytu obou forem P. teres v průběhu vegetace.
