    The PPV genome region spanning C-terminal of NIb gene, coat protein (CP) and 3'-non translated region (NCR) gene of a challenging PPV isolate Greek interacting with in transgenic plum has characterized. Different typing assays including ELISA, RT-PCR, RLFP and sequencing (Cter-NIb, CP and 3'NCR region) reveals that challenging PPV-Greek belongs to a typical M subgroup. High sequence (both nucleotide 'nt' and amino acid 'aa') homology were found with M type isolates SK68 (nt 99.0% and aa 100%) or PS (nt98.1% and aa 99.3%) and with recombinant isolate BOR 3 (nt 96.0% and aa 97.4%). The PPV-Greek detected in transgenic plum, resistance clone C-5 or susceptible clone C-6 has shown a few mutations, at the position nt 8441 of Cter-NIb and nt 9179 of CP genein C-6 and at the position nt 9068 in C-5. The mutation in CP gene seems to drive from transgenic transcript, suggest the evidence of template switching between CP cistron of challenging virus and homologous transgenic transcript. These
