Analysis of multiple virus-infected grapevine plant reveals persistence but uneven virus distribution
Original language description
The LN33 grapevine plant was artificially infected with Grapevine leafroll-associated virus-1 (GLRaV-1), Grapevine virus A (GVA), Grapevine virus B (GVB), Rupestris stem pitting-associated virus (RSPaV), and an unclassified tymovirus. Although infected with GVB, corky bark symptoms never appeared, and this virus had no destructive effects on the LN33 grapevines under our experimental conditions. Dormant canes, sampled before the beginning of the vegetation period, were used as suitable targets to detectmentioned viruses using both the ELISA and RT-PCR method. For GLRaV-1, the ELISA- based diagnosis unexpectedly appeared more effective than RT-PCR; probably due to the insufficient specificity of the primers used, not reflecting the actual genetic variability of the virus. Distribution of viruses in long-term infected grapevine plants showed different degrees of irregularity, depending on the individual viruses.
