    Detection of translocation rob(1;29) in bull sperm using a specific DNA probe

    The Robertsonian translocation rob(1;29), connected with reduced fertility, is widespread in different cattle breeds. After laser microdissection, DOP-PCR, cloning and sequencing, a highly sensitive translocation-specific DNA probe, suitable for detection of rob(1;29) in cattle cells, including spermatozoa was designed. One thousand sperm from each of five heterozygous translocation carrier bulls were analyzed using this probe and a control probe for chromosome 6. Signals of the translocation-specific probe were detected in 48.8%, 50.9%, 50.1%, 51.8%, and 54.8% of spermatozoa, respectively. Semen samples from five control bulls showed only signals of the control probe for chromosome 6. No sperm head with signal of the translocation-specific probe was observed among 1000 spermatozoa from a chimeric (XX/XY) bull, showing 57.5% of 59,XX,rob(1;29) and 42.5% of 60,XY cells in cultured peripheral lymphocytes, demonstrating that female cells do not pass through the process of spermatogenesis.

    Robertsonská translokace rob(1;29), spojená s redukcí fertility je široce rozšířená u různých plemen skotu. Laserovou mikrodisekcí, DOP-PCR, klonováním a sekvenováním byla získána vysoce citlivá sonda specifická pro tuto translokaci, která je využitelnápro detekci rob(1;29) v buňkách skotu včetně spermií. S touto sondou a kontrolní sondou pro chromosom 6 bylo analyzováno 1000 spermií od každého z 5 heterozygotních býků - nositelů rob(1;29). Signál rob(1;29)-specifické sondy byl detekován v 48.8%, 50.9%, 50.1%, 51.8%, a 54.8% spermií. Vzorky semene 5 kontrolních býků vykazovaly pouze signál sondy pro chromosom 6. V 1000 spermiích chimerického býka s karyotypem 59,XX,rob(1;29) (57.5%)/ 60,XY (42.5%) v periferních lymfocytech nebyl pozorován signál rob(1;29)-specifické sondy, což demonstruje, že samičí buňky neprocházejí procesem spermatogeneze.
