In this study, we develop the HPLC assay for the direct determination of alpha-tocopheryl succinate (TOS) entrapped in the liposomes as well as monitoring of TOS chemical stability in the liposomal lyophilisates during 6-month storage period. The composition of mobile phase was optimised for separation of TOS, alpha-tocopherol (TOH) as TOS degradation product and alpha-tocopheryl acetate as internal standard. The quantification of TOS in liposomal formulation and liposomal sediment enables to calculateentrappment efficiency of TOS (EETOS). For the liposomal lyophilisates stored during 6 months, the obtained values of EETOS were higher than 90%. The chemical stability of liposomal entrapped TOS was investigated by monitoring the level of TOH. Generally, the content of liposomal TOH did not exceed 5% of the liposomal TOS content.
