    Construction of surface-modi?ed metallochelating microbubbles (MBs) was proved using recombinant green ?uorescent protein (rGFP) with 6 HisTags. MBs composed of DPPC with various contents of metallochelating lipid DOGS-NTA-Ni were prepared by intensive shaking of liposome suspension under atmosphere of sulfur hexa?uoride by 3M ESPE CapMix device. Static light scattering, ?ow cytometry and optical microscopy were used to determine mean size (2.1-2.8 um) and concentration of MBs (109/mL). MBs were lyophilized in the presence of sucrose. These MBs were stable in vitro (half-live of MBs in bovine serum at 37 °C was 3-7 min) and in vivo (mouse model). DPPC-based metallochelating MBs provided clear and very contrast image of the ventricular cavity in mouse heart soon after injection as observed by ultrasonic system. Commercial MBs (Bracco) were used as standard. Site selective and stable binding of rGFP-HisTag onto the surface of metallochelating MBs was observed by confocal microscopy.
