Sperm plasma membrane integrity is commonly evaluated by intravital staining tests or by a test of membrane tolerance and resistance to hypo-osmotic conditions (HOS test). We proved that despite good sperm viability as detected by a simple HOS test, thedegree of membrane resistance can be low. The difference in sperm survival rates during the 120-min test between groups of boar ejaculates containing higher and lower percentages of HOS-positive and eosin-negative sperm was statistically significant eventhough the other parameters of sperm analysis did not significantly differ. Moreover a higher percentage of sperm was recovered after thawing of bull ejaculates with a higher percentage of HOS-positive and eosin-negative sperm. The HOS test in combination with intravital staining is a procedure providing additional information concerning dynamic changes in the membrane resistance. It allows assessment of semen quality from the perspective of its further use, e.g. cryopreservation.
