Ischemic stroke is leading cause of mortality and morbidity with serious socioeconomic impact. The occlusion causing the stroke is usually formed by a clot. Unfortunately nowadays method are not able to imagine it. The most common method of imaging CTA shows brain arteries with flow and arteries without flow are missing. The better imaging methods can lead to more precise treatment such as better targeting of stent retrievers as well as improvement in diagnosis of other thromboembolic events like pulmonary embolism and leg ischemia. As a first step to achieve patients’ clot imaging we employed in vitro models. Simple static models were used for fast screening. Sophisticated flow models mimicking average size of middle cerebral artery were used for selected binders targeting fibrin. In vitro models to simulate ischemic stroke were prepared from tubes or by lost element method with use of 3D printing and polydimethyl siloxan. Models were connected to a pump to simulate blood flow. Clots were prepared from citrated human plasma coagulated with thrombin at room temperature. The clot was inserted to the model to simulate complete occlusion. After that fibrin targeting peptide with contrast agent was injected. 30 min later, the clot was extracted and prepared for confocal microscopy. Microscopy results show that the binder binds to fibrin.
