Exosomes can mediate intercellular communication during tumor development and are enriched in nucleic acids, proteins and lipids (e.g. sphingolipids, ceramides and cholesterols). Lipids are a heterogeneous group of substances in terms of structure and function. The subject of this study was glycosphingolipids, which have a sugar residue attached to a ceramide in their structure. Deregulation of lipid metabolism is an important alteration in cancer. The current project describes changes in glycosphingolipids in exosomes. First of all, the spectrum of glycosphingolipids were compared in tumor and nontumor (T/N) cell lines with aim to find diagnostic markers of cancer cells. Our results showed that tumor cells had increased glycosphingolipids GD2 and GM1. Moreover, UGCG and B4GalT5 were also increased. Next, exosomes were isolated from paired patient samples by ultracentrifugation. The size, polydispersity and number of particles were examined and compared between T/N samples. Dynamic light scattering analysis showed that both more exosome populations were distributed in tumor samples (higher polydispersity) and the size of tumor exosomes was bigger. Changes in exosomal glycosphingolipids were analyzed by flow cytometry and electron microscopy. In conclusion, our results showed that exosomes isolated from tumor samples were polydisperse and had an altered glycosphingolipid profile. For the first time, we described the methodology for the isolation of exosomes from paired samples of T/N tissue obtained from one patient. Our results confirmed the role of glycosphingolipids in exosomes as potential diagnostic biomarkers in cancer patients.
