Original language description
Spirochetal genus Treponema includes several closely related pathogenic spirochetes. Treponema pallidum ssp. pallidum (TPA) is the causative agent of syphilis, T. pallidum ssp. pertenue (TPE) causes yaws. Although these two subspecies are microscopicallyand serologically indistinguishable, they cause different diseases. Little is known about genome differences between TPA and TPE genomes. The aim of this work was to employ dideoxyterminator sequencing for sequence differences discovery between Nichols(TPA) and Samoa D (TPE) genomes. All heterologous regions were successfully subjected to DDT sequencing and consensus sequences were obtained using three different approaches: 1) direct PCR product sequencing, 2) primer walking and 3) small insert library sequencing. Identified sequence differences will be used together with CGS data to compile the complete genome sequence of Samoa D strain.
