    Association of matrix metalloproteinases gene variants with recurrent aphthous stomatitis

    Background: Recurrent aphthous stomatitis (RAS) is a common inflammatory disease of oral mucosa manifested by erosions and ulcers. Variability in genes coding matrix metalloproteinases (MMPs), the proteolytic enzymes modulating inflammation, has been previously associated with RAS. The aim of our study was to investigate six single nucleotide polymorphisms (SNPs) in genes for gelatinases (MMP2, MMP9) and stromelysin (MMP3) in patients with RAS and healthy controls. Subjects and methods: Totally, 226 subjects were enrolled in this case-control study: 71 patients with RAS and 155 healthy individuals were genotyped for SNPs in the MMP2 (rs243865, rs2287074), MMP3 (rs679620, rs35068180) and MMP9 (rs3918242, rs17576) genes by TaqMan qPCR or by PCR with restriction analysis. Results: While allele frequencies of rs3918242 MMP9 polymorphism and haplotype MMP9 (rs3918242, rs17576) were marginally significantly different between patients with RAS and healthy controls (P=0.04 and P=0.06, respectively), distribution of allele, genotype and haplotype frequencies of the other studied SNPs were found similar in both groups. Nevertheless, the period of ulcer healing significantly less frequently exceeded one week in the MMP3 (rs35068180) 6A/6A carriers and MMP3 (rs679620) CC carriers compared to other genotypes (P=0.01 for both). Conclusions: Our study suggests that MMP9 gene variability, affecting MMP9 expression and activity, may be involved in the development of RAS in the Czech population. Moreover, MMP3 variants could influence the duration of ulcer healing.
