    Glycosyltransferase Fold Recognition Analysis of the Mycobacterium Tuberculosis Genome Sequence

    There is very little knowledge about enzymes involved in the biosynthesis of unique and unusual cell wall structure of M. tuberculosis, the leading cause of human death. The genome M. tuberculosis contains 3,951 protein-coding sequences. Putative function of some of them was predicted using a combination of sequence alignment and motif comparison [1]. Using this approach, only a small number of putative GTs was identified. Recent crystal structure determination of GTs demonstrated that they adopt a limited number of topology that can be characterized using fold recognition approach [2]. We therefore proposed first to focus on 3D-topology identification of potential GTs in M. tuberculosis and second to apply this method to the complete amino acid sequences encoded by the M. tuberculosis (http://genolist.pasteur.fr/TubercuList/). The ProCeryon program that is based on Knowledge-Based Potentials [3] was used with parameters optimised on existing structures of GTs. This approach allows to
