Thirteen years ago a new application for the evaluation of enzymatic reactions in capillary electrophoresis was proposed and developed, electrophoretically mediated microanalysis (EMMA). In this method the capillary is used not only as a separation medium but also as a reaction chamber. Since its discovery by Bao and Regnier [1], the EMMA methodology has been utilized in a number of biochemical systems [2,3]. In this work, the EMMA method was applied to determine cyanide after its on-capillary enzymaticreaction with rhodanese (thiosulfate: cyanide sulfur transferase, EC 2.8.1.1).Consequnetly cyanide thus could be determine as thiocyanate with adequate sensitivity by means of CE with simple UV detection. Compared to other assays, the EMMA method is rapid, can be automated, and requires only small amount of samples.
