    Study of cryptogein recognition in plant defense reaction.

    Cryptogein is proteinaceus elicitor isolated from Phytophthora cryptogea able to trigger a plant defense reaction. We prepared a series of cryptogein mutants for study of interaction cryptogein - receptor. Cryptogein mutants were tested for ability to induce plant defense early events, i.e. synthesis of active oxygen species, pH changes and NO production, in suspension tobacco cells. Moreover interaction between cryptogein or cryptogein mutants and receptor was studied by piezoelectric biosensor. We focused on study of cryptogein mutants with low affinity to tobacco receptor. These mutants showed high threshold concentration in triggering production of active oxygen species, pH changes and NO production. However sensitivity of defense reactions was different between the mutants. Low affinity of cryptogein mutants to receptor indicate that mutation affect the binding site of cryptogein. Further, by piezoelectric biosensors was characterized interaction of cryptogein and tobacco receptor

    Cryptogein je proteinový elicitor isolovaný z Phytophthora cryptogea, který je schopný vyvolat obrannou reakci u rostlin. Připravily jsme řadu mutantů cryptogeinu pro studium interakcí s jeho receptorem. Tyto mutanty byly testovány při vyvolání rané fázeobranné reakce (syntéza aktivních forem kyslíku, produkce NO) u tabákových buněk. Dále byla studována přímá interakce těchto mutantů s receptorem pomocí piezoelektrického biosenzoru. Získali jsme řadu mutantů crytogeinu se sníženou schopností vyvolat obrannou reakci u rostlin. Tato práce by měla přispět k hlubšímu určení vazebných míst pro cryptogein u rostlin tabáku.
