Amino acids (AA) have no native fluorescence thus chemical modification (so called derivatization) of AA has to be performed prior the detection by a fluorescent detector. There is a lot of derivatization reagents commercially available differing in reactivity, excitation and emission wavelengths, quantum yield, product stability etc. Naphtalene-2,3-dicarbaldehyde (NDA) is a non-fluorescent reagent giving fluorescent product of primary amines. Derivatization procedure is quite simple, however has to beoptimized to minimize undesired by-products of benzoin condensation. The by-products can exhibit fluorescence or form crystals which can clog capillary. The novel derivatization protocol using NDA for AA derivatization is subject of this contribution.
