TPA is known activator of protein kinase C. This activation subsequently reduces proliferation and induces differentiation and apoptosis of human myeloid leukaemia cell line HL-60. We have studied possible role of arachidonic acid metabolites in these processes. The reduction of proliferation and the induction of differentiation were not influenced either by indomethacin or MK-886. While indomethacin did not affect the percentage of apoptotic cells, MK-886 combined with TPA significantly enhanced the amount of apoptotic cells in concentration in which MK-886 alone did not induce apoptosis. Assuming that the enhancement of apoptosis by MK-886 was caused by 5-LPO inhibition, blocking the 5-LPO activity could contribute to leukaemia treatment.
