Cultivation method and polymerase chain reaction coupled with immunomagnetic separation (IMS-CM and IMS-PCR) are new technoques used for the detection of bacteria in foodstuffs. In this study, Salmonella was isolated from various real food samples by magnetic Dynabeads M-280 anti-Salmonella using both IMS-CM and IMS-PCR. Food samples included (milk, eggs, meat products, confectionery, vegetable, fruit products) and total 408 specimens were investigated. 49 samples (i.e. 12%) were found Salmonella positive by IMS-CM. Bacteria coupled on immunomagnetic particles were also used in IMS-PCR. Growing atypically on sensitive medium two Salmonella strains were identified using IMS-PCR. False negative results were obtained by conbventional technques. The abovementioned results demonstrated that IMS-CM and IMS-PCR techniques are more sensetive in real food samples analysis than conventional cultivation procedures.
