Original language description
Tuberculosis is one of the most difficultly treated and in many cases lethal infectious disease caused by Mycobacterium tuberculosis. This pathogen is characterized by unique cell envelope - crucial factor of virulence. Mycobacterial glycosyltransferases(GTs) are enzymes playing crucial role in synthesis of mycobacterial cell wall. GT Rv3782 synthesizes mycobacterial galactan - very important element of the cell wall and its lack can cause growth inhibition up to death. Therefore GT Rv3782 is very attractive target for development of new drugs against tuberculosis. Study of GTs is very difficult because these enzymes are unstable and often membrane-associated. Vector pMTW is a new expression system for expression of problematic proteins. pMTW was madeas a combination of two vectors pMAL and pTWIN. The basis of vector is maltose-binding protein from pMAL enabling correct protein folding and one or both inteins from pTWIN vector for easy removal of fusion protein.
