Hairy root (HR) tissue culture is an efficient system for studying gene function and hormone responses in Brassicaceae species, including Brassica napus and Brassica rapa. HR transformation enables rapid generation of transgenic root lines, allowing functional analysis of introduced genes. In this study, cytokinin and auxin reporter systems were used to monitor hormonal responses in HRs. A cytokinin-responsive reporter (TCSv2:3×VENUS) showed increased signal intensity after cytokinin treatment, confirming active cytokinin signaling. Additionally, auxin output reporters (DR5 and pIAA motif-based constructs) demonstrated auxin responsiveness under different treatment conditions. These systems allow visualization of hormone activity in root tissues with high spatial resolution. HR-derived plants can be regenerated and used for further genetic and functional analyses. Furthermore, the expression of TAA1 gene family members in B. napus was investigated, revealing differential expression patterns. Overall, HR tissue culture provides a versatile platform for studying hormonal regulation and gene expression in Brassicaceae.
