In a group of 18 strains (15 bacteria and 3 yeasts), their production of nanoparticles of elemental selenium (SeNPs) after selenization was studied. The strains were previously selenized by cultivation in the media containing 10 mg/L of sodium selenite, for 24 h under optimal conditions for each strain. Following that, SeNPs were determined by inductively coupled plasma mass spectrometry (ICP-MS) in single particle measurement mode (sp-ICP-MS), where the dispersion of NPs is introduced into ICP-MS, and the signal of individual NPs is detected in a time-resolved record. The diameter and numerical concentration of NPs was then calculated based on NPs signal intensities and number of signals, respectively. The accuracy of NPs size determination by sp-ICP-MS was also verified by transmission electron microscopy (TEM). The crystal structure and shape of the NPs were measured by TEM. Furthermore, the composition of the nanoparticles was studied using an energy dispersive X-ray spectrometer (EDX) connected to the TEM. In most of the tested strains, the presence of SeNPs was demonstrated, which in many samples formed the majority (4-68 % of the total selenium content). The mean diameter of the SeNPs ranged from 80 to 260 nm, with the cocci clearly producing larger NPs (130-260 nm) than yeasts, lactobacilli and bifidobacteria (80-135 nm). However, the particle size distributions were wide in many cases and the sizes of the individual SeNPs ranged from 50 to 400 nm. Except for the yeast Pichia fermentans CCDM 2012, where SeNPs were produced intracellularly, and for Saccharomyces cerevisiae CCDM 272 and Lacticaseibacillus rhamnosus CCDM 146 with SeNPs produced both intracellularly and extracellularly, extracellular SeNPs were observed in all other strains. All particles had the structure of amorphous Se, except those produced by the Lacticaseibacillus rhamnosus CCDM 146 strain, in which the crystalline structure of trigonal Se was confirmed. The only strain in which the formation of SeNPs was not confirmed by either TEM or sp-ICP-MS is Lacticaseibacillus paracasei CCDM 213.
