    Molecular Phylogenetic Assessment of Fungal Biodiversity in Olomouc Region Soil: An ITS-Based Approach

    The advent of modern molecular marker technologies has greatly facilitated the rapid identification, characterization, and detection of important soil fungi, such as species of Fusarium, which were previously difficult to detect using traditional microbiological methods. Molecular tools can now rapidly characterize fungal biodiversity using small quantities of DNA or direct fungal mycelium samples. Various molecular markers, including RFLP, RAPD, SSR, AFLP, ITS, and IGS, have accelerated the identification and characterization of soil fungi over recent decades. Among these, the internal transcribed spacer (ITS) region of ribosomal RNA genes is widely used due to its ability to identify fungi at the genus and species levels, supported by its extensive representation in sequence databases such as NCBI. This study, ITS sequencing of fungal species has expanded the understanding of soil fungal populations in the Olomouc region, paving the way for broader fungal diversity assessments using samples from different ecosystems in future studies.
