Comparison of the performance of two different newly designed real-time PCR assays applicable for S. aureus/ MRSA identification

  • Original language description

The environment of farm animals was identified as an important source of pathogenic S. aureus strains including the antibiotic-resistant forms. These could contaminate the food chain or infect the humans. In order to improve the quality of existing microbial diagnostic tools, we designed two real-time PCR assays for identification of Staphylococcus aureus/ MRSA strains. The methods worked with different platform of PCR product detection: SYBR green dye and dual-labelled hydrolysis (TaqMan) probes. We compared the performance of the both assays with regard to its practical application. The both PCR assays featured with reproducible and robust amplification. The limit of the detection between the assays differed significantly. The SYBR green assay enabled detection of tens of S. aureus genome copies unlike hundreds of genome copies at TaqMan assay. A group of thirty reference strains of S. aureus/ MRSA and twenty-eight strains of different Staphylococcus spp. were analysed using the opti
