The aim of our work was to provide a new, high-throughput and reliable method intended for rapid identification of MRSA in raw cow milk. The duplex real-time PCR assay working with the dual-labelled hydrolysis probes was developed. The sequence specificity of the nuc gene was utilized to design S. aureus specific assay. Detection of the methicillin resistance was based on the mecA gene sequence identifying. A collection of twenty S. aureus/ MRSA reference strains and twenty non - S. aureus strains inclusive Staphylococcus spp. with potential occurrence in milk was analysed to validate the parameters of the method. The incubation of artificially contaminated milk samples in solution of Bird-Parker medium was employed as a selective pre-enrichment step.The PCR assay featured with robust, specific and reproducible amplification.
