Original language description
The necrotic tissue emerging in certain types of wounds is necessary to remove in the first wound healing phase. Removing of this tissue is called the debridement. Very painful surgical debridement can be replaced by painless enzyme debridement if we solve the problem with very low enzyme stability. For this reason, the proteolytic enzymes were immobilized on the surface of silica nanofibers. Silica nanofibers were produced by electrospinning from free liquid surface and then modify by silanization reaction. Three silanes were tested for silanization (3-Aminopropyl triethoxysilane, 3-Glycidyloxypropyl trimethoxysilane and 3-Mercaptopropyl trimethoxysilane). A two-step method with succinic anhydride and N-Hydroxysuccinimide ester was used for functionalization of the silica nanofibers. After this procedure, protease from Bacillus licheniformis, trypsin from hog pankreas and bromelain from pineapple stem were immobilized on functionalized silica nanofibers. The proteolytic activity of immobilized enzymes was tested in dependence on pH value and temperature. The most dependent on pH was bromelain from pineapple stem and the most dependent on temperature was protease from Bacillus licheniformis. As a comparative method for functionalizing the surface of nanofibres, a well-known method with glutaraldehyde was used. The proteolytic activity of immobilized enzymes with succinic anhydride and N-hydroxysuccinimide ester as a ligands was comparable or higher than the proteolytic activity of immobilized enzymes using glutaraldehyde as a ligand.
